摘    要:目的探讨自杀基因HSV-TK对肺癌细胞增殖活性影响.方法利用嗜银染色AgNOR观察肺癌细胞系Anip973在经阳离子脂质体介导的自杀基因系统HSV-TK转染后应用阿昔洛韦(ACV),观察细胞形态并计数,观察其增殖活性的变化.结果经AgNOR嗜银染色72h测定,其结果表明,G、G-Lipo、G-P细胞AgNOR平均数无明显差别,而G-pTK细胞的AgNOR平均数明显低于前三种细胞,具有显著差异.结论自杀基因HSV-TK/GCV系统对肺癌细胞株Anip973的细胞增殖活性具有抑制作用,对其临床应用具有理论意义.

Abstract:Objective To evaluate the th e proliferative activity influence of HSV-TK/ACV transfected by cationic liposom e to lung cancer cell. Methods This study was carried out by silver staining to lung cell line Anip 973 whcih was tranfected HSV-TK gene and treated with ACV, then count AgNOR. Results After 72 h silver staining, AgNOR were counted, the average number of G?G-Lipo?G-p wer e no significan difference and the average AgNOR number of the cell which was tr ansfected TK gene was significantly low to the others. Conclusion The suicide gene HSV-TK can inhibit the proliferative activity of lung cancer cell line Anip 973, and it is promising to be used in clinical genetherap y.
