运动预处理对力竭运动诱导的大鼠大脑皮质细胞凋亡的影响

摘    要:目的:探讨运动预处理对力竭运动诱导的大鼠大脑皮质细胞凋亡的保护作用。方法:40只雄性SD大鼠随机分为对照组(C组,n=8),力竭运动组(EE组,n=16),运动预处理组(EP组,n=16)。EP组进行持续4周的中等负荷游泳训练(无负重游泳,60 min/d,6 d/w)后,EE组和EP组负重3%体重进行一次性力竭游泳运动。HE染色观察神经细胞形态学变化,免疫组织化学法和蛋白免疫印迹方法检测各组大鼠大脑皮质Bcl-2和Bax蛋白表达。结果:EE组大鼠大脑皮质神经细胞凋亡较EP组增多;EP组大鼠大脑皮质Bcl-2阳性表达强度较EE组升高,EP组大鼠大脑皮质Bax阳性表达强度较EE组下降。结论:运动预处理能够减少力竭运动诱导的大鼠大脑皮质细胞凋亡从而对大脑皮质产生保护作用;运动预处理可能通过影响Bcl-2、Bax蛋白表达从而对细胞凋亡起调控作用。

Abstract:Objective To investigate the effect of exercise preconditioning on exhaustive exercise-induced apoptosis of neurons in the brain cortex of rats.Methods Forty SD rats were divided into three groups at random:control group(C,n = 8),exhaustive exercise group(EE,n = 16)and exercise preconditioning group(EP,n = 16).Group EP carried out moderate exercise preconditioning for 4 weeks(swimming with non-weight bearing,60 min/d,6 days/wk),and group EE and group EP underwent an exhaustive swimming carrying 3% self body-weight objects.Change in brain cortex morphology was observed by HE staining,and the Bcl-2 and Bax proteins in brain were examined by immunohistochemistry and Western blot.Results Apoptosis index of brain cortex neuron was higher in group EE than that in group EP.Protein expression of Bcl-2 was higher in group EP and protein expression of Bax was higher in group EE.Conclusion Moderate exercise preconditioning can reduce the apoptosis of the brain cortex neuron cells induced by exhaustive exercise through affecting the Bcl-2 and Bax protein.
