雌激素对阿尔茨海默病大鼠模型MAPK蛋白表达的影响

摘    要:目的探讨雌激素对阿尔茨海默病(AD)大鼠模型学习记忆能力及丝裂霉原活化的蛋白激酶(MAPK)蛋白表达的影响。方法30只大鼠行卵巢切除术(OVX)后,均分成OVX组和雌激素替代治疗组(ERT),然后采用1μlofAβ1~40(10μg/μl)立体定位单侧海马内注射建立AD动物模型,通过水迷宫实验检测动物模型的学习、记忆能力;免疫组化检测MAPK蛋白表达情况。结果ERT组AD动物模型的水迷宫逃避潜伏期较OVX组明显缩短(P<0.05),同时ERT组AD动物模型的海马CA1、CA3区的MAPK表达上调(P<0.05)。结论雌激素可以改善AD动物模型的认知功能,其神经保护作用可能与上调海马MAPK表达有关。

Abstract:Objective To investigate if estrogen modulates the learning and memory and expression of mitogen- activated protein kinase (MAPK) in model rats of Alzheimer's disease (AD). Methods Ovariectomy (OVX) was performed in 30 rats that were divided averagely into OVX group and estrogen replacement therapy (ERT) group. AD rat model was established by injection of amyloid-beta peptide 1 μ L of A β 1 - 40(10 μg/μL) into the hippocampus in 30 rats. Learning and memory ability was determined by Morris water maze, the expression of MAPK was assessed by immunohistochemistry method. Results The escape latency of Morris water maze was decreased in ERT group compared with the OVX group (P〈0.05), the expression of MAPK was higher at CA1, CA3 area of hippocampus in ERT group than in OVX group (P〈0.05). Conclusion ERT improves the cognitive deficits of AD rats, which may be related to the increased expression of MAPK in hippocampus.
