肝细胞生长因子促血管内皮细胞增殖及迁移的研究

摘    要:目的:探讨重组人肝细胞生长因子(rhHGF)对人脐静脉血管内皮细胞(HUVEC)增殖和迁移的影响及其促血管生成作用.方法:从脐静脉体外分离培养HUVEC,采用四甲基偶氮唑蓝法(MTT)观察rhHGF对HUVEC的增殖作用,倒置显微镜观察HUVEC的迁移,透射电镜观察超微结构的改变及流式细胞仪检测细胞周期的改变.结果:(1)rhHGF对培养的HU-VEC有显著的促增殖和迁移作用,且成浓度和时间依赖性.(2)rhHGF促使HUVEC细胞超微结构的改变,线粒体和粗面内质网增多,常染色质增多(3)细胞周期分布改变:S期、G2/M期细胞增多.结论:rhHGF能显著促进HUVEC的增殖和迁移,提示其可能具有促血管生成作用.

Abstract:Objective:To investigate the effects of hepatocyte growth factor on proliferation and migration of human umbilical vein endothelial cells,and to further study the angiogenesis of HGF.Methods:The cultured endothelial cells in vitro were isolated from human umbilical vein.The proliferation of HUVECs were measered by MTT and their migration were observed using reverse microscope,The changes of celluar ultrastructure were observed under electron microscope and flow cytometry was applied to observe the effects of rhHGF on cell cycle.Results:rhHGF could promote the proliferation and migration of HUVECs dose-dependently and time-dependently.There were obvious changes in celluar structure,rhHGF could increase mitochondrion,rough endoplasmic reticulum and euchromatin.Conclusion:rhHGF could promote the proliferation and migration of HUVECs significantly,which suggests that HGF might have the effect of angiogenesis.
