葛根素对小鼠子宫ERα、ERβ蛋白表达的影响

【目的】研究葛根素对小鼠子宫ERα、ERβ蛋白表达的影响。【方法】成年雌性小鼠腹腔注射给予葛根素500mg／ks和50mg／kg7d后，采用免疫组织化学染色结合图像分析手段检测子宫组织ERα、ERβ蛋白免疫反应阳性细胞数及其面积。【结果】与对照组相比，葛根素500mg／kg组E邴阳性细胞数和阳性细胞面积显著高于对照组（P〈0．01），ERα则无差异；葛根素50mg／kg组ERa和E邸阳性细胞数及阳性细胞面积均无差异（P〉0．05）。己烯雌酚组ERα和ERβ阳性细胞数和阳性细胞面积均显著高于对照组（P〈O．01）。【结论】500mg／kg葛根素明显增强子宫组织ERβ蛋白表达，而对ERα蛋白无明显影响，己烯雌酚则对两种ER亚型的表达均有显著上调作用，提示葛根素对子宫内膜的刺激增殖作用远低于雌激素的原因可能与其对ERβ的选择性上调有关。

Effects of puerarin on estrogen receptor alpha and beta proteins in mouse uterus

Objective ] To study the effects of puerarin on the estrogen receptor alpha and beta proteins in mouse uterus. Methods] infra perifoneally into puerarin were given matured female mice in dose of 500 mg/kg or 50 mg/kg every day. After 7 days, ERalpha and ERbeta proteins in uterus were determined with immunohistochemical mothod. The numbers and areas of receptors- positive cells in the endometrium were determined by image analytic system. Results] After treated with puerarin of 500 mg/kg for 7 days, when the numbers and areas of receptors - positive cells of ERbeta protein were signifigantly increased in the endometrium （ P 〈 0.01）, no obvious difference was observed in ERalpha （ P 〉 0.05）, no statistic differences were determined in ERalpha and ERbeta proteins in puerarin of 50 mg/kg （ P 〉 0.05）. But in diethylstilbestrol group, the numbers and area of receptor- positive cells of ERalpha and ERbeta proteins were signifigantly increased in the endometrium （ P 〈 0.01 ）, Condusions] The expression of ERβ protein in endometrium is significantly improved by puerarin of 500 mg/kg, but no significant effect on ERαprotein is observed. Diethylstilbestrol up - regulated significantly the expressions of both ERa and ERβ proteins. It is possible that the weaker stimulat on to endometrinm of puerarin than estrogen relates to its selective up - regulating ERβ.
