海藻酸钠/壳聚糖的免疫隔离及生物相容性

摘    要:目的:探讨微囊材料海藻酸钠/壳聚糖的生物相容性和免疫隔离作用。方法:以海藻酸钠-壳聚糖-海藻酸钠(ACA)为包囊,分别将微囊化PC12细胞、裸PC12细胞、空微囊移植入帕金森病(Parkinson′s disease,PD)模型鼠纹状体内,以阿朴吗啡检测移植前后大鼠旋转行为的改变。HE染色后观察纹状体病理形态学的变化。对移植的微囊化PC12细胞回收后再培养,以MTT和台盼兰检测细胞活性。结果:微囊化PC12细胞移植能够改善PD模型鼠的旋转行为,与移植前和空囊移植组相比差异有显著性(P<0.05),包囊无破裂,无肿瘤形成,囊周炎症反应轻微。裸PC12细胞移植组大鼠的旋转行为改善仅持续了2个月,且部分大鼠颅内有致死性肿瘤形成。回收微囊化PC12细胞再培养生长良好,MTT法和台盼兰法检测显示细胞具有生物活性。结论:海藻酸钠壳/聚糖具有良好的生物相容性,ACA微胶囊能够起到有效的免疫隔离和抑制肿瘤形成的作用。

Abstract:Objective To investigate the immunoisolation effect and biocompatibility of sodium alginate/chitosan. Methods The alginate chitosan alginate (ACA) was used as the microcapsule.Encapsulated PC12 cells,PC12 cells and empty microcapsule were transplanted respectively into the striatum of the Parkinsonian rats.The behavioral changes were tested by apomorphine.The morphological alterations were observed by striatal HE stain. The retrieved encapsulated PC12 cells were recultured and their activities were tested by MTT and Trypan blue test. Results The encapsulated PC12 cells could improve the rotational behavior of Parkinson′s disease (PD) model rats and there was a significant difference ( P <0 05) comparing with pretransplantation group and empty capsule transplantation group. There was no disruption and no tumor formation in this group, and the inflammatory reaction around the capsule was slight. Although the unencapsulated PC12 cells could also improve the rotational behavior,the symptom improvement lasted only two months.In addition,several of the grafts developed into mortal tumor. The well growth and the good biological activities of the retrieved PC12 cells were proved using the MTT and Trypan blue test. Conclusion Sodium alginte/chitosan has a good biocompatibility. ACA microcapsule can both isolate the host immune system effectively and prevent the hypersegmentation of the cells inside the capsule.
