摘    要:目的 探讨白细胞介素17(IL-17)及其受体调控酪氨酸激酶(JAK)/转录激活因子-3(STAT3)信号通路促进喉癌血管生成的作用. 方法 收集喉癌手术切除标本70 例,癌组织通过不同病理分级,采取70 例喉癌患者的外周血. 采用ELISA 法检测外周血中 IL-17、血管内皮生长因子 A ( VEGF-A)的浓度,分析其与临床病理特征的关系. Western blot 法检测喉癌组织中IL-17、IL-17受体( IL-17R)、JAK1/2、p-JAK1/2、STAT3、p-STAT3、VEGF-A、环氧合酶-2 ( COX-2 )、基质金属蛋白酶-9(MMP-9)等信号分子水平的变化. 结果喉癌患者血清中IL-17、VEGF-A的含量与喉癌的分化程度、淋巴结有无转移和TNM分期有关( P<0. 05 ) , Western blot结果显示在喉癌发展的不同时期,随着喉癌的恶化, IL-17、IL-17R的表达逐渐增强,相关信号通路分子JAK1/2、p-JAK1/2、STAT3、p-STAT3、VEGF-A、COX-2、MMP-9 表达也显著增加. 结论 IL-17可能调控JAK/STAT3信号通路,在喉癌的血管生成中起到重要的作用.

The role of IL-17 and its receptor in invasion and metastasis of laryngeal cancer
Abstract:Objective To investigate IL-17 and its receptor regulate JAK/STAT3 signaling pathway in the angio-genesis of laryngeal cancer. Methods Serum levels of IL-17 and VEGF-A were determined in 70 laryngeal carci-noma patients using ELISA. The levels as well as clinicopathological features were analyzed. Western blot assay was used to detect the expressions of IL-17,IL-17R,JAK1/2,p-JAK1/2,STAT3,p-STAT3,VEGF-A,COX-2,MMP-9 in 70 cases of laryngeal carcinoma. Statistics methods were applied to analyze the results. Results Serum levels of both IL-17 and VEGF-A levels in patients were related to the degree of differentiation of laryngeal cancer, lymph node metastasis and TNM stage (P<0. 05), were unrelated to age,sex,primary tumor site. Western blot analysis showed that at different times in development of laryngeal cancer, with the deterioration of laryngeal cancer, IL-17, IL-17R expression gradually increased, JAK1/2,p-JAK1/2,STAT3,p-STAT3,VEGF-A,COX-2 and MMP-9 ex-pressions were also significantly increased. Conclusion IL-17 may play an important role in angiogenesis of laryn-geal cancer via regulating JAK/STAT3 signaling pathway.
