超氧阴离子自由基对NIH3T3细胞的转化作用

摘    要:首次选取NIH3T3细胞,对比检测外源性O^-2和致癌,促癌剂争生转化的能力。结果表明,O^-2单独作用组较正常对照组和启动剂量B(a)P单独作用组之转化率提高了近3倍;当O^-2与启动剂量B(a)P协同作用时,转化率上述两组分别提高约11倍和10倍;而用SOD和CAT清除胞外O^-2时,转化率下降82%。

Abstract:he transformation frequency(TF)of N IH3T3 cells was examined to study the effect ofextracellular superoxide anion(O2)on cell transformation,The TF in cell cultures treated withO2 alone was 3 times greater than that in control group(A) or that in cultures received initiatingdose of B(a)P alone(B).In cultures treated with TPA in combination with B(a)P,the TF wasabout 20 and l8 times greater than that in group A and B respectively.When and B(a) Pacted cooperately,TF was nearly 11 and 10 times greater than that in group A and B respective-ly.Superoxide dismutase(SOD)in combination with catalase(CAT),as scavengers of activeoxygen,reduced the TF by 82%,These results show that can act as a complete carcinogenespecially as a promoter.
