燕麦β-葡聚糖对糖尿病大鼠胰岛功能的影响

摘    要:目的:探讨燕麦β-葡聚糖(Oglu)对四氧嘧啶致糖尿病大鼠(DM)胰岛功能的影响。方法:大鼠腹腔注射四氧嘧啶建立DM模型后灌胃Oglu,剂量200mg/kgbw,连续14d,实验结束后测定大鼠血清胰岛素和C肽含量,并取大鼠胰腺,观测胰岛细胞组织形态学变化、分析胰岛β细胞凋亡和p53、bcl-2基因表达。结果:经Oglu治疗后,DM大鼠血清胰岛素含量(29.34±5.1)和C肽含量(1.89±0.28)较治疗前(16.8±4.9和1.32±0.31)明显增高(P<0.01),胰腺细胞团由1~2个增加到2~4个,且受损胰岛细胞有明显好转迹象;胰岛bcl-2基因表达增强,p53基因表达受抑,同时胰岛β细胞的凋亡进程得到缓解。结论:Oglu对大鼠受损的胰岛细胞有一定修复作用,可促进bcl-2和抑制p53基因表达。

Abstract:Objective: To study the effect of oat glucan on pancreatic function in diabetic rats. Method:The diabetic rats induced by alloxan were given oatβ-glucan (200 mg/kg bw) for 14 d and then the serum insulin , C peptides were determined. The morphology and apoptosis of β cells, and expression of p53 and bcl-2 genes were observed. Results: By giving oatβ-glucan , the serum insulin and C peptide were increased significantly in diabetic rats (P<0.01), and the pancreatic cells began to restore. The expression of bcl-2 genes were boosted up and the p53 genes were repressed. Conclusion: Oatβ-glucan could restore the function of βcells. The mechanism was associated with inhibition of p53 gene and enhanced expression of bcl-2 gene.
