| 摘 要： | 目的探讨金黄色葡萄球菌(SAU)对人脐静脉内皮细胞(HUVEC)的黏附、内化及诱导细胞凋亡的发生、发展的机制和致病意义。方法常规方法制备SAU悬液,分别感染HUVEC细胞30 min、1、24、h。采用光学显微镜和荧光显微镜观察SAU对HUVEC的黏附、内化情况,采用流式细胞仪、DAPI荧光染色及DNA琼脂糖凝胶电泳分析SAU对HUVEC细胞诱导凋亡的影响。结果SAU与HUVEC共育30 min发现有细菌黏附细胞表面,1 h后有少量细菌侵入胞内,并伴有凋亡现象的发生,随着时间的延长,内化的细菌数没有增加,凋亡现象通过流式细胞仪、DAPI荧光染色及DNA琼脂糖凝胶电泳分析明显的被观测到。结论SAU通过黏附、内化侵入内皮细胞并诱导内皮细胞发生凋亡。 |
