创伤患者外周血淋巴细胞自噬变化及其与糖代谢的关系

摘    要:目的 观察创伤患者外周血淋巴细胞的自噬变化规律以及与创伤后血精变化的关系.方法 根据损伤严重度评分(ISS)或简明损伤定级标准(MS)将创伤患者分为轻伤组、重伤组、严重伤组,同时设立健康对照组,分别于创伤后1、3、5、10 d采集外周血,采用密度梯度离心结合贴壁分离法分离创伤患者外周血淋巴细胞,应用激光共聚焦显微镜和透射电镜观察细胞自噬现象;将分离得到的淋巴细胞进行MDC染色并裂解,应用荧光分光光度计(激发光波长380 nm、发射光波长525 nm)测定荧光强度进行定量分析;于创伤后1、3、5 d检测患者血糖水平,分析创伤后自噬与血糖的相关性.结果 与健康对照组比较,创伤患者外周血淋巴细胞自噬水平明显升高,有显著变化(P<0.01).结论 创伤后外周血淋巴细胞自噬水平与创伤严重程度、创伤后血糖成正相关.

A study on the change of autophagy in peripheral blood lymphocytes following trauma
Abstract:Objective To investigate the change rule of autophagy in peripheral blood lympho-cytes in trauma patients and its relationship with the change of blood sugar after trauma. Methods Ac-cording to ISS or AIS, all the patients were divided into three groups : mild, moderate, and severe trauma groups. The control group was estabhshed. The peripheral blood of three trauma groups was collected at the day 1,3,5 and 10 after trauma, and lymphocytes were obtained by density gradient centrifugation combined with adherence separation. The autophagy phenomenon was observed by using laser confocal microscopy and transmission electron microscope. The lymphocytes were stained with MDC, and lysed for quantitative detection of the fluorescent intensity by fluorospectrophotometer (wavelength of excitation light was 380 nm,and that of emission light was 525 run). The blood sugar levels were determined at the 1st, 3rd, and 5th day respectively after trauma, and the correlation between autophagy and blood sugar levels was ana-lyzed. Results Compared with control group,the level of autophagy in lymphocytes in trauma patients was significantly increased in trauma groups (P < 0.01). Conclusion The level of autophagy in lymphocytes after trauma had a positive correlation with the trauma degree and the blood sugar.
