摘    要:目的:研究Pax9基因在斑马鱼早期发育阶段腮弓中的表达。方法:利用RT-PCR技术直接克隆Pax9特异性基因片段,制作针对Pax9的基因探针,选取斑马鱼发育早期多个时段的胚胎进行整胚原位杂交。结果:成功克隆Pax9基因,合成针对Pax9的基因探针,发现Pax9基因在受精16h的斑马鱼胚胎中开始有特异性信号出现,还可以明显地看到腮弓上的Pax9基因特异性信号在24~72h发育过程中的迁徙过程。结论:初步了解了Pax9基因在斑马鱼早期发育阶段的表达情况。

Abstract:Objective:To study the expression of Pax9 gene in the early development stage of zebrafish branchial arch.Methods:Synthesized gene probes for Pax9 by cloning specific gene fragment of Pax9 gene with RT-PCR directly were used to carry out the whole mount in situ hybridization with embryo from different stages during early dental development.Results:With synthesis gene probes for Pax9 by clone Pax9,we found that specific signals of Pax9 gene began to appear in zebrafish embryos 16h after fertilization,and we could see the whole migratory process of Pax9 specific signals in branchial arch during 24h-72h of development clearly.Conclusion:The expression of Pax9 gene during the early stage of zebrafish was shown successfully.
