人酸性成纤维细胞生长因子支持原代培养下丘脑神经元的存活和突起生长

摘    要:初步研究了人酸性成纤维细胞生长因子(aFGF)对无血清培养条件下新生大鼠下丘脑神经元的突起生长和促生存作用。结果可见,在此培养物中加入纯化的aFGF后,神经元的存活数增加,神经突起生长,数目增多,aFGF的这种作用在一定范围内呈剂量效应关系。实验还表明,aFGF可增加神经元胞体直径和体积,流式细胞分析亦表明神经元蛋白总量提高。本结果提示,纯化的aFGF对在神经内分泌中占重要地位的下丘脑也有神经营养作用。其作用机制尚有待进一步研究。

Abstract:The effects of human acidic fibroblast growth factor (aFGF) on the survival andneurite growth of hypothalamic neuron were observed in the cell culture with serum free medium.The results showed an increase both in the number of surviving cells and in the outgrowth ofneurites. In the presence of aFGF, the diameter and volume of neuron as well as the content ofprotein in the cells were markedly increased on image analysis and FACS. All effects were closelycorrelated with the dose. The results suggest that aFGF may play a neurotropic role in culturedhypothalamic neuron.
