人冷冻精液巴氏染色法的应用评价

摘    要:目的:探讨人冷冻精液应用巴氏染色法的染色效果。方法:30例人精液标本冷冻保存后,冷冻精液涂片应用巴氏(Papanicolaou)染色法进行染色,观察精子和非精子细胞的染色效果,检测冷冻精子畸形率。应用透射电镜观察冷冻精子头部(n=8)超微结构的改变。结果:冷冻保存后的精子畸形率,与冷冻前的比较无统计学差异(P>0.05)。冷冻精子头、颈、尾部,各种形态类型精子以及非精子细胞的着色效果与新鲜精液的对比无明显不同。冷冻精液涂片后自然干燥所需的时间较长。透射电镜观察到冷冻精子头部浆膜及顶体发生肿胀和破损。结论:巴氏染色法可以应用于冷冻精液的涂片染色,实际应用时需注意冷冻精液含有甘油等的特点。

Abstract:Objective: To evaluate the efficacy of the Papanicolaou staining method on cryopreserved human semen. Methods: Thirty human semen samples were enrolled into this study. The Papanicolaou staining method was used to stain cryopreserved semen. Sperm morphology was determined by light microscopy. Sperm head ultra- structure was observed by transmission electron microscopy (n=8). Results: There was no significant difference in abnormal sperm morphology rate between fresh and cryopreserved sperm (P>0.05). No obvious difference was observed on efficacy for Papanicolaou-stained cryopreserved sperm and other cells compared with the fresh ones. It took much long time to allow the smear of cryopreserved semen to dry. Ultrastructural observation showed that sperm plasma membrane and acrosome membranes were swollen, interrupted, or damaged. Conclusion: The Papanicolaou staining method can be used on cryopreserved human semen. It gives good staining of cryopreserved sperm and other cells.
