组织生物样本库中冷冻组织质量鉴定体系的建立

目的：建立组织生物样本库中储存的冷冻组织样本的质量鉴定体系。方法：随机选取浙江省台州医院组织库中肝癌、胃癌冷冻组织及正常对照组织各20例,HE染色评估标本取材的准确性,Agilent2100生物分析仪检测RNA纯度和完整性,Western blot和琼脂糖电泳分别测定组织样本的蛋白质和DNA质量。结果：HE染色观察到受检样本肿瘤部分均达到80%以上,RNA完整系数（RIN）为7.1～8.4,28s/18s比值为1.5～1.8,受检样本蛋白质及DNA均保存完好,无明显降解。结论：本研究介绍的冷冻组织质量鉴定体系,采用的方法可靠,可以作为组织生物样本库冷冻组织样本质量鉴定的有效且常规方法。

Establishment quality of assessment system for human frozen tissues from the tissue biobank

Objective：To establish a standard protocol for quality assessment of human frozen tissues from the tissue biobank.Methods：Twenty frozen tissues and normal control tissues of hepatocellular carcinoma and gastric cancer were respectively randomly selected and embedded,sectioned,stained with HE and pathologically checked.Agilent 2100 bioanalyzer was used to check the RNA quality.Western blot and agarose electrophoresis were used to detect the quality of the protein and DNA from these frozen tissues.Results：HE staining showed that all the detected samples contained 80% or greater tumor.RNA integrity number（RIN） and 28 s/18 s ratio were respectively 7.1～ 8.4 and 1.5～1.8.The good quality of the protein and DNA from the randomly selected frozen tissues was observed.Conclusion：The pipeline for quality assessment introduced in this paper is appli-cable as a routine method in quality control of human frozen tissues from the tissue biobank.
