异丙酚对大鼠局灶性脑缺血再灌注后HSP70mRNA和HSP70蛋白表达的影响

摘    要:目的研究异丙酚对大鼠局灶性脑缺血再灌注后HSP(热休克蛋白)70mRNA和HSP70蛋白的动态表达规律及其作用的影响。方法采用线栓法建立大鼠局灶性脑缺血再灌注模型,应用原位杂交原理检测HSP70 mRNA,应用组织化学染色法检测HSP70蛋白。结果异丙酚使HSP70 mRNA的表达增强,时间延长(P<0·05);HSP70蛋白表达量显著增多(P<0·05)。结论异丙酚能显著促进脑缺血再灌注时HSP70的表达。提示异丙酚一方面发挥自身的神经保护作用,另一方面也可以通过诱导HSP70的合成而发挥神经保护作用。

Abstract:Objective To investigate the effects of propofol on expression of HSP70mRNA and HSP70 in focal cerebral ischemia and reperfusion and to discuss its probable mechanism. Methods Experimental animals and group division are the same as part one. Use in situ hybridization and immunochemistry to test expression of HSP70mRNA and the HSP70 at different time points in focal cerebral ischemia and reperfusion. Results Propofol can enhance the expression of HSP70mRNA and prolong the duretion of high level HSP70mRNA(P<0.05); Expression of HSP70 of propofol group is significantly more than that of ischemia and reperfusion group(P<0.05). Conclusion Focal crebral ischemia can induce a series of stress reaction, induce the expression of HSP70 in brain tissue,protect the injured neurons and enhance the tolerance of brain tissue to ischemia and anoxia. Propofol can enhance the expression of HSP70 arfter ischermia and protect neutrons from being damaged.
