烧伤大鼠血清对骨髓红系粒系造血的影响

摘    要:目的:观察烧伤血清对骨髓红系粒系造血的影响,探讨促进造血细胞增殖的物质。方法:用5kW溴钨灯致大鼠背部30%体表面积Ⅲ度烧伤,伤后无菌抽取大鼠血清。按10mg/L蛋白加入到骨髓红系或粒系体系中培养。结果:烧伤后3,12,24,48,72和96h,骨髓红系(CFU-E,BFU-E)和粒分(CFU-GM)集落数均显著高于正常对照组;伤后24h达337.7,179.7,240.5个/1×105骨髓有核细胞(bonemarrowcell,BMC);对照组仅98.5,68.7,105.2个/1×105BMC;差异非常显著(t=8.31,9.65,7.84,P<0.01)。结论:烧伤血清对骨髓红系、粒系造血均有明显的促进作用。

Abstract:AIM:To observe the influence of blood serum from burned rats on the bone marrow erythrocyte/granulocyte progenitor cells so as to explore the materials in burn serum facilitates haematopoietic cells proliferation. METHODS:Rats were inflicted with 30% TBSA III degree burn with 5 kW bromo tungsten lamps. The blood serum from these animals was collected after injury. Then it was added to the culture medium of erythrocyte progenitor cells or granulocyte macrophage progenitor cells at final concentration of 10 mg/L. RESULTS:The colony numbers of colony forming unit (CFU E,BFU E) and colony forming unit giant mitochondria (CFU GM) formed after addition of the blood serum from rats with burn was significantly higher than that from normal rats at 3, 12, 24, 48, 72 and 96 hours after injury and reached its peak value at 24 hours after injury. The colony numbers of bone marrow cell (BMC)reached (337.7,179.7, and 240.5/105 )after burn, which were obvious from control groups(98.5, 68.7 and 105.2/105). The difference was obvious (t=8.31,9.65, 7.84,P< 0.01).
