骨髓瘤细胞对骨髓间充质干细胞成骨分化潜能的影响

摘    要:目的检测骨髓瘤细胞对骨髓间充质干细胞成骨潜能的影响,探讨骨髓瘤细胞在骨髓瘤骨病发病机制中的作用。方法分离培养骨髓间充质干细胞,鉴定其生物学特性。采用midiMACs阳性免疫磁珠分选系统分离多发性骨髓瘤(MM)患者骨髓CD138+细胞,与骨髓间充质干细胞行Transwell成骨共培养,两周后进行定时定量PCR检测间充质干细胞成骨指标骨特异性碱性磷酸酶(ALP)和Cbfa1的mRNA表达量,Von Kossa染色鉴定其钙质沉积程度改变。结果骨髓CD138+细胞可以抑制骨髓间充质干细胞ALP和Cbfa1的mRNA表达量,间充质干细胞钙质沉积也相应减少。结论骨髓瘤细胞可抑制骨髓间充质干细胞的成骨潜能,这种抑制作用可能参与了骨髓瘤骨病的发生。

Abstract:Objective To study the effect of myeloma cells on the osteogenic potential of bone marrow mesenchymal stem cells (MSCs), so as to explore the role of myeloma cells in the myeloma bone disease mechanism. Methods Biological characters of bone marrow MSCs were studied. CD138^+ myeloma cells were isolated using midiMACs positive magnetic microbeads sorting system and cocuhured with MSCs in transwell systems in osteogenie medium for two weeks, and then real-time RT-PCR and Von Kossa staining were employed to detect the changes of osteogenic markers and mineral deposition of MSCs. Results Myeloma cells inhibited osteogenic markers and mineral deposition of marrow MSCs. Conclusion Myeloma cells inhibit osteogenic potential of marrow MSCs, which may participate in the mechanism of myeloma bone disease.
