苦丁茶总皂苷的水提工艺研究

摘    要:目的优化苦丁茶中主要活性成分总皂苷的水提工艺条件,保证了主要活性成分的充分溶出。方法以苦丁茶皂苷E为对照品,香草醛-高氯酸法显色,用紫外分光光度法在550 nm处测定总皂苷的含量。采用L9(34)正交试验,以苦丁茶总皂苷含量及浸膏得率为指标,确定合理的水提工艺。结果最佳提取工艺条件为15倍量水,每次1 h,直火回流提取3次。结论该工艺重复性好,对苦丁茶水提部分下一步的制剂工艺研究有很好的指导作用。

Abstract:Objective To optimize the water process conditions of the main active ingredients of Kudingcha,and ensure dissolving it sufficiently.Methods A colorimetric method was established in 550nm with Kudingoside E as the index and Vanilla aldehyde-high chlorine acid as the coloring reagent.Designed L9(34) orthogonal test with the ratio of total saponins and condensate to determine the rational water extraction technology.Results The best extract condition was with 15 times quantity water to extract 3 times,and 2h each time.Conclusion This process has good repeatability,is a good guiding role for the preparation process of Kudingcha’s water extraction.
