茶多酚对2型糖尿病患者外周血早期内皮祖细胞衰老及氧自由基损伤的影响

摘    要:目的:通过体外诱导培养2型糖尿病(T2DM)患者外周血早期内皮祖细胞(earlyEPC),观察茶多酚(TP)对其衰老和氧自由基损伤的影响。方法:分别采集20例T2DM患者及15例正常对照人群外周血各20ml,获取单个核细胞后,在包被有纤维连接蛋白(FN)的培养板中加入含有血管内皮生长因子(VEGF)等的M199培养基,诱导earlyEPC分化,于第6天加入不同浓度TP干预24h后,测定earlyEPC衰老情况及丙二醛(MDA)含量。结果:两组样本分别给予TP(1mg/L、10mg/L、100mg/L)干预后,earlyEPC的衰老细胞明显减少(P<0.01),MDA含量明显降低(P<0.05),但TP对实验组earlyEPC的衰老和MDA改善情况不如对照组明显(P<0.05);100mg/L的TP对T2DM患者MDA的改善效果不明显(P>0.05)。结论:适宜浓度TP能够不同程度改善T2DM患者earlyEPC的衰老及氧自由基损伤。
