乳腺癌患者血浆微泡BCRP mRNA水平的改变及其临床分析

摘    要:目的:探讨血浆微泡乳腺癌耐药蛋白(BCRP)mRNA水平与乳腺癌化疗疗效及乳腺癌组织相关基因的关系。方法选择接受新辅助化疗的乳腺癌患者60例作为乳腺癌组,选择同时期健康体检者60例作为对照组。检测血浆微泡BCRP mRNA转录水平以及乳腺癌组织相关基因中凋亡基因Caspase-3、Caspase-9和增殖相关基因Survivin、Bcl-2 mRNA含量。结果乳腺癌组患者血浆微泡中BCRP mRNA含量高于对照组,差异有统计学意义(t=31.289,P﹤0.05);CR/PR组患者血浆微泡中BCRP mRNA含量低于SD/PD组,差异具有统计学意义(P﹤0.05);乳腺癌组织中凋亡基因Caspase-3、Caspase-9 mRNA含量低于癌旁正常组织,增殖相关基因Survivin、Bcl-2 mRNA含量高于癌旁正常组织,差异有统计学意义(P﹤0.05)。结论血浆微泡中BCRP的mRNA含量与caspase-3、caspase-9 mRNA含量呈负相关,与Survivin、Bcl-2 mRNA含量呈正相关。血浆微泡中BCRP mRNA含量可作为乳腺癌辅助化疗疗效的预测指标,为乳腺癌化疗提供指导依据。

Abstract:Objective To investigate the relationship between the breast cancer resistance protein (BCRP) mRNA lev-el in plasma microvesicles and efficacy of chemotherapy, and related genes. Method 60 cases of breast cancer patients who were given neoadjuvant chemotherapy were included as breast cancer group, while another 60 cases of healthy adults were enrolled as control group. The BCRP mRNA level in plasma microvesicles and the level of apoptosis related breast cancer genes, such as caspase-3, caspase-9 and proliferation related genes, including Survivin, Bcl-2 were ana-lyzed. Result The BCRP mRNA level in plasma microvesicles were higher in breast cancer group as compared with con-trol group, with significant difference observed (t=31.289, P<0.05);While the levels in CR/PR patients were lower in that of SD/PD patients, which were significantly different (P<0.05);For apoptosis genes, the mRNA of caspase-3 and caspase-9 were less in breast cancer tissues than in the adjacent normal tissues, while for the proliferation related genes, the mRNA of Survivin and Bcl-2 were higher in breast cancer tissues than in the adjacent normal tissues, and all differenc-es were statistically significant (P<0.05). Conclusion The mRNA level of BCRP in plasma microvesicles was negatively correlated with the mRNA levels of caspase-3 and caspase-9, while was positively correlated with the mRNA level of Survivin and Bcl-2. The BCRP mRNA level in plasma microvesicles is predictive for the efficacy of adjuvant chemothera-py in breast cancer, and is useful in guiding the chemotherapy for breast cancer.
