脑缺血再灌注大鼠额叶皮质内Bcl-2和HSP70的表达及川芎嗪、尼莫地平的干预作用

摘    要:为了探讨脑缺血再灌注(CIR)大鼠额叶皮质细胞内Bcl-2和HSP70的表达及川芎嗪、尼莫地平的干预作用,本研究采用Bannister’s颈动脉血引流法复制Wistar大鼠CIR模型,将动物随机分成6组,即正常对照组、假手术组、模型组、川芎嗪治疗组、尼莫地平治疗组和川芎嗪+尼莫地平联合治疗组,每组各15只动物。川芎嗪治疗组按100mg/kg经腹腔注射,尼莫地平组经静脉给药(1mg/kg),联合治疗组则同时给上述剂量药物。三个治疗组均自缺血起给药一次,然后每24h给药一次。所有动物额叶皮质的切片均采用免疫组织化学技术检测Bcl-2和HSP70阳性细胞的表达情况。结果显示:正常对照组Bcl-2和HSP70无阳性表达,假手术组仅HSP70有少量表达,模型组Bcl-2和HSP70阳性细胞数明显有所增多,而川芎嗪或尼莫地平治疗组Bcl-2和HSP70阳性细胞数明显多于模型组(P<0.05)。与单独用药组相比,川芎嗪+尼莫地平联合治疗组Bcl-2、HSP70阳性细胞数明显增多(P<0.05)。以上结果表明川芎秦和尼莫地平能够上调CIR时Bcl-2和HSP70的表达,提示它们可能在CIR中发挥保护作用。

Abstract:To identify the expression of Bcl-2,HSP-70 and interference effects of ligustrazine or/and nimodipine on cerebral ischemia reperfusion (CIR) cortex of frontal lobe in rats,the Bannister's drainage method in neck artery was used to make CIR model of Wister rats. Then the rats were randomly divided into 6 groups:normal contral group,sham operated group,model group,ligustrazine treatment group,nimodipine treatment group and the treatment group of ligustrazine with nimodipine,and each group 15 rats. The lingustrazine (100 mg/kg) was injected into abdominal cavity in ligustrazine treatment group. The nimodipine (1 mg/kg) was injected by intravenous injection in nimodipine treatment group. The same drug dose was injected in combined treatment group. At the same time from ischemia,drugs were given at the first time,then every 24 h once. The sections of cortex of frontal lobe of all rats were detected for Bcl-2 and HSP70-immunoreacitivity in the neuronal cell bodies by immunohistochemical method. The results showed that no Bcl-2 and HSP70 immunopositive neurons were observed in control group,but a little Bcl-2 and HSP70 immunopositive neurons were found in sham operated group. The number of Bcl-2 and HSP70 positive neurons increased in model group. In ligustrazine or nimiodipine treatment group,the number of Bcl-2 and HSP70 positive neurons was significantly higher than that in the model group (P<0.05). Compared with alone ligustrazine or nimiodipine treatment group,the number of Bcl-2 and HSP70 positive neurons in combing ligustrazine with nimodipine group was significantly higher (P<0.05). The above results suggest that ligustrazine and nimiodipine may upregulate the expression of Bcl-2 and HSP70 in CIR cerebral cortex,which indicates that they might play a protecting role for CIR.
