细胞焦亡与骨质疏松症关系研究

摘    要:骨质疏松症(osteoporosis, OP) 是一种以骨组织微结构退化、骨密度和骨量降低为特征的骨代谢疾病。近年来国内外相关实验研究总结OP以及骨细胞病理进程发现,细胞焦亡(pyroptosis)机制对该病有着重要介导作用。细胞焦亡属于机体的天然免疫反应,多种炎症小体介导的细胞焦亡在OP中发挥重要作用,但细胞焦亡介导OP的发生机制尚未明确。因此,阐明细胞焦亡的信号机制和与OP的联系,从而采取针对性的靶向治疗,对于防治OP有着重要意义。此外,近年来随着中医药分子水平的研究逐渐深入,中医药抑制细胞焦亡改善OP骨细胞代谢的研究逐渐增多,中医药参与靶向治疗有望成为临床上治疗该病的突破方向。本文将综述细胞焦亡机制与OP的相关性研究,分析细胞焦亡途径影响骨质疏松的病理机制,阐述细胞焦亡调控OP中骨细胞代谢和平衡的具体机理,为细胞焦亡介导骨质疏松症的发病机制研究提供科学依据,并且为寻找潜在的靶点提供思路。

Research progress on the relationship between pyroptosis and osteoporosis
Abstract:Osteoporosis (OP) is a bone metabolic disease characterized by bone microstructure degradation, bone mineral density and bone mass reduction. In recent years, relevant experimental studies in China and abroad have summarized OP and the pathological process of bone cells.Pyroptosis after activation resists local inflammation, which belongs to the natural immune response of the body. Pyroptosis mediated by a variety of inflammatory bodies play an essential role in OP. However, the mechanism of pyroptosis-mediated OP has not yet been clarified.Therefore, it is significant for the prevention and treatment of OP to clarify the signaling mechanism of pyroptosis and its relationship with OP so as to adopt targeted therapy. In addition, in recent years, with the gradual deepening of the research on the molecular level of traditional Chinese medicine, the study of traditional Chinese medicine inhibiting pyroptosis and improving the metabolism of OP bone cells has gradually increased. Traditional Chinese medicine participating in targeted therapy may become a breakthrough direction in clinical treatment of the disease. This article reviews the correlation between pyroptosis mechanism and OP, analyzes the pathological mechanism of pyroptosis pathway affecting osteoporosis, and expounds the specific mechanism of pyroptosis regulating the metabolism and balance of bone cells in OP, which provides a scientific basis for the research on the pathogenesis of pyroptosis-mediated osteoporosis, and provides ideas for finding potential targets.
