甲泼尼龙对体外培养许旺细胞分泌神经生长因子的研究

摘    要:目的研究甲泼尼龙对体外培养许旺细胞分泌神经生长因子的促进作用。方法体外培养兔许旺细胞,纯化后的细胞配成单细胞悬液,按5×10^4个/孔的细胞数量接种于预先铺过明胶的4孔培养板中。根据加入药物浓度随即分成四组:A组,空白对照组;B组,加入0.1mg/L甲泼尼龙;C组,加入1.0mg/L甲泼尼龙;D组,加入5.0mg/L甲泼尼龙。分别于加药后第2、4、6、8、10天吸取培养液做NGF、CNTF、GDNF定量测定。结果①B、C、D组许旺细胞生长速度明显快于A组,三种神经生长因子分泌量明显多于A组,差异有统计学意义(P〈0.01),B、C、D组之间差异无统计学意义(P〉0.05);②NGF的检测结果有优于CNTF、GDNF的趋势(P〈0.01),CNTF和GDNF的促分泌情况差异无统计学意义(P〉0.05)。结论甲泼尼龙能促进体外培养许旺细胞的增殖和神经生长因子的分泌。小剂量应用和大剂量应用差异无统计学意义。

Abstract:Objective To study the methylprednisolone on cultured Schwann cells secrete nerve growth factor to promote the role. Methods The purified Schwann cells match single - cell suspension, according to a 5 × 10^4/hole against the number of cells in the pre - shop gelatin over the 4 - hole plate in training. According to the accession to the concentration of the drug was divided into four groups: A group, control group; B group, adding 0.1 mg/ L methylprednisolone; C group, adding 1.0 mg/ L methylprednisolone ; D group, to join 5.0 mg/ L methylprednisolone. Dosing in the first 2,4,6,8,10 d learned to do medium NGF, CNTF, GDNF quantitative analysis. Results B, C, D groups Schwann cells faster than the growth rate of group A, three kinds of nerve growth factor secreted significantly more than group A, statistically significant differences (P 〈 0.01 ), B, C, D group between the secretion was no significant difference between each other (P 〉 0. 05). Conclusions Methylprednisolone can promote the in vitro proliferation of Schwann cells and nerve growth factor secretion. Application of low - dose and high dose had no significant differences in the application.
