EphA2基因在大肠腺癌组织中的表达

目的探讨EphA2基因在大肠腺癌组织中表达的意义.方法采用免疫组化（SP）技术对154例大肠腺癌组织和相应正常大肠黏膜组织中EphA2蛋白表达进行检测;采用逆转录-聚合酶链反应（RT PCR）技术对66例大肠腺癌手术切除新鲜癌组织和相应正常大肠黏膜组织中EphA2 mRNA的表达进行检测.结果EphA2蛋白表达在154例癌组织中明显高于正常大肠黏膜组织（P＜0.001）,且EphA2蛋白的高表达与肿瘤生长方式、组织学分级、浸润深度相关（P＜0.05）.66例大肠腺癌组织中22例EphA2 mRNA呈阳性表达,正常黏膜组织中21例EphA2 mRNA呈阳性表达,两者相比差异无统计学意义（P＞0.05）,EphA2 mRNA的表达与肿瘤患者性别、肿瘤部位、生长方式、体积、组织学分级和肿瘤转移均无相关性（P＞0.05）;但是与肿瘤患者的发病年龄和肿瘤的浸润深度相关（P＜0.05）.结论EphA2基因与大肠腺癌的发生、肿瘤细胞的迁移和浸润能力相关.

The expression of erythropoitin-producing hepatocellular A-2 gene in colorectal adenocarcinoma and its impact on DNA ploid types and cell proliferation

Objective To investigate the significance of erythropoitin-producing hepatocellular (Eph)A2 expression in colorectal adenocarcinoma.Methods One hundred and fifty four paried tissue specimens of coloreetal adenocarcinoma and corresponding normal mucosa were examined for EphA2 pro tein expressions by immunohistochemistry(SP method).Of 66 paried specimens obtained front surgical resection,the expression of EphA2 mRNA was detected by RT-PCR.Results EphA2 protein expres sions in colorectal adenocarcinoma was significantly increased tban that in normal mucosa(P<0.001), and it related to the growth pattern,grade and invasion depth of carcinoma(P<0.05).There was no statistical difference of EphA2 mRNA expression between colorectal adenocarcinoma(n=22)and normal mucosa (n=21)(P>0.05)and it was also not correlated with gender and tumor site,growth pattern and metastasis(P>0.05),but correlated with age and invasion depth of tumor(P<0.05). Conclusions EphA2 gene had impact on tumorigenesis,migration and invasion of colorectal adenocarcinoma.
