槲皮素阻断热休克蛋白70表达对艾灸预处理抑制胃黏膜损伤细胞凋亡的影响

摘    要:目的通过槲皮素灌胃阻断热休克蛋白(HSP)70表达,观察艾灸预处理对大鼠急性胃黏膜损伤指数及细胞凋亡的影响,以进一步探明艾灸的细胞保护作用及其与HSP70的关系。方法40只大鼠随机分为捆绑对照组(A组)、模型组(B组)、艾灸+模型组(C组)、槲皮素+艾灸+模型组(D组)、槲皮素+模型组(E组)。D、E组每日采用槲皮素灌胃。C、D组每日艾灸足三里、梁门,以上处理共8 d。第9日,B、C、D、E组均采用无水乙醇灌胃制作大鼠急性胃黏膜损伤模型。采用western-blot方法检测胃黏膜HSP70蛋白表达;TUNEL法检测胃黏膜细胞凋亡指数(AI)。结果与A组比较,模型组大鼠胃黏膜AI明显增高(P〈0.05),胃黏膜HSP70表达升高(P〈0.01)。与B组相比,C组大鼠AI明显降低(P〈0.05),HSP70表达显著升高(P〈0.01);D组大鼠胃黏膜AI值明显增加(P〈0.01),HSP70表达未见上调。与C组相比,D组大鼠胃黏膜AI值升高明显(P〈0.01),HSP70表达明显下调。结论艾灸预处理上调胃黏膜HSP70表达,急性病胃黏膜损伤所致的细胞凋亡被抑制。槲皮素阻断HSP70合成后,艾灸预处理抑制急性胃黏膜...

Effect of Quercetin Blocking HSP70 Expression on Pre-Moxibustion Suppressing Gastric Mucosal Cell Apoptosis

Abstract:Objective To observe the effects of pre-moxibustion on gastric mucosal ulcer apoptosis index(AI) and cell apoptosis in acute gastric mucosal injuring rats by perfusing quercetin into the stomach to blocking HSP70 expression,to further prove up the cell protecting effect of moxibustion and its connection with HSP 70.Methods Forty healthy Wistar rats were randomly assigned to group A(binding control),group B(model),group C(moxibustion + model),group D(quercetin + moxibustion + model),and group E(quercetin + model). Quercetin was given by gavage in group D and E, and moxibustion at Zusanli and Liangmen acupoint in group C and D every day for 8 days. At the ninth day, acute gastric mucosal injuring model was made by ethanol in group B, C, D and E. AI was checked by TUNEL method, and HSP70 by western-blot method in gastric mucosa of all groups. Results Compared with group A, the value of AI was increased obviously (P 〈0.05), and HSP70 expression of gastric mucosa was up-regulated in group B (P〈0.01). Compared with group B, the value of AI was reduced obviously (P〈0.05) and HSP70 expression of gastric mucosa was up-regulated obviously in group C (P〈0.01). The value of AI was increased obviously in group D (P〈0.01), but HSP70 expression of gastric mucosa has no change. Compared with group C, the values of AI was raised obviously, but HSP70 expression of gastric mucosa was down-regulated markedly in group D. Conclusion While HSP70 expression of gastric mucosa was up-regulated by treatment with pre-moxibustion, the cell apoptosis was suppressed in acute gastric mucosal injuring rats. But after HSP70 expression was blocked by quercetin, the effect of restraining the cell apoptosis with pre-moxibustion was canceled. This study certificates that pre-moxibustion has the gastric mucosal protecting efficacy, and the effect is related with up-regulation of HSP70 expression.
