摘    要:目的探讨间充质干细胞(MSCs)对人前列腺癌细胞上皮-间质转化(EMT)和侵袭能力的影响。方法以干扰素γ(IFNγ)预处理的间充质干细胞[MSCs(IFNγ)]的培养上清处理人前列腺癌PC-3细胞,观察PC-3细胞形态的变化,并以划痕实验、Transwell实验检测PC-3细胞的迁移、侵袭能力,使用实时定量聚合酶联反应(qRT-PCR)法检测EMT相关的基因E-cadherin、Vimentin、N-cadherin的mRNA表达情况。结果与PBS预处理的对照组相比,实验组的PC-3细胞有从上皮的类圆形细胞向间质的梭形细胞转化的趋势;与对照组相比,实验组中E-cadherin mRNA表达下调,而Vimentin及N-cadherin mRNA表达上调(P均〈0.05);实验组PC-3细胞的迁移和侵袭能力均强于对照组(P〈0.05)。结论 IFNγ预处理的MSCs可促进前列腺癌细胞发生EMT,并使得前列腺癌细胞迁移、侵袭能力增强。

Abstract:Objective To explore the effect of mesenchymal stem cells( MSCs) on epithelial-mesenchymal transition( EMT) and invasion in human prostate cancer cells. Methods MSCs were pretreated with interferon γ( IFNγ). The human prostate cancer PC-3 cells were further pretreated with the cultured supernatants of these cells.The morphological change of PC-3 cells was observed. The wound-healing and Transwell experiments were performed to detect migration and invasion of PC-3 cells,while real-time qRT-PCR assay was used to detect the mRNA expression of EMT-related genes E-cadherin,Vimentin and N-cadherin. Results Compared with control group pretreated with PBS,PC-3 cells in the experimental group had the trend to transform from round epithelial cells to fusiform mesenchymal cells. Compared with control group,the expression of E-cadherin mRNA downregulated significantly in the experimental group. On the contrary,the expression of Vimentin and N-cadherin mRNA upregulated remarkably( all P 〈 0. 05). The capacity of migration and invasion of PC-3 cells in the experimental group was significantly stronger than that in the control group( P 〈 0. 05). Conclusion MSCs pretreated with IFNγ could promote the EMT of prostate cancer cells and enhance the invasion and metastasis.
