磷脂酰肌醇3-激酶/蛋白激酶B信号通路对生长激素介导的心肌成纤维细胞增殖的影响

摘    要:目的探讨生长激素(GH)对心肌成纤维细胞(CFs)增殖的影响及其作用机制。方法以不同浓度的重组人生长激素(rhGH)(100、200、500、1 000 ng/ml)和磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/Akt)抑制剂LY294002(10μmol/L)单独或同时作用CFs 48 h后,采用MTT法检测细胞增殖,Wertern Blot检测PI3K/Akt信号途径中Akt和磷酸化Akt(p-Akt)的蛋白水平的变化。结果 rhGH呈浓度依赖性的促进CFs的增殖;rhGH增加p-Akt的蛋白水平;LY294002可阻断rhGH诱导的CFs的增殖。结论生长激素可通过激活PI3K/Akt信号通路促进心肌成纤维细胞的增殖。

Abstract:Objective To investigate the effect of growth hormone( GH) on the proliferation of cardiac fibroblasts( CFs) and its mechanism.Methods CFs were treated by different concentrations of recombinant human growth hormone( rhGH)( 100,200,500,1000 ng/ml) with or without phosphatidylinositol-3-kinase/protein kinase B( PI3K/Akt)inhibitor LY294002( 10 μmol /L) for 48 hours. MTT assay was used to determine the proliferation of CFs. Western Blot was used to detect the protein levels of Akt and phosphorylated Akt( p-Akt) in PI3 K /Akt signaling pathway. Results rhGH promoted the proliferation of CFs in a concentration-dependent manner. rhGH increased the protein level of p-Akt. LY294002 could depress the rhGH-induced proliferation of CFs. Conclusion GH promotes the proliferation of CFs by activation of PI3 K /Akt signaling pathway.
