摘    要:目的:筛选并优化复盆调经颗粒的制备工艺。方法:通过药效学实验比较和确定提取纯化工艺路线,正交设计优化制备工艺条件。结果:水提醇沉提取物的主要药效学作用与传统水提工艺提取物比较无显著差异,但能显著增强缩宫素引起的离体大鼠子宫平滑肌的作用(P<0.01);能显著增加未成年雌性小鼠阴道开放例数(P<0.01),能显著增加去卵巢小鼠的子宫重量百分率(P<0.01),能显著增加PMSG处理后小鼠的排卵数(P<0.01);通过正交试验优化确定的水提醇沉工艺条件为:药材加10倍量水提取2次,每次1h,合并滤液减压浓缩至相对密度为1.10(55℃),加乙醇使药液含醇量达75%,静置24h,浓缩干燥。结论:水提醇沉工艺能除去杂质,减小复盆调经颗粒的服用剂量,并保证了原处方的疗效;优化得到的工艺简单合理、稳定可行。

Abstract:Objective:To screen and optimize the preparation techniques of Fupen-tiaojing granules.Methods:The extraction and purifying route was screened by using pharmacodynamic experiment and the preparation conditions were optimized by orthogonal design.Results:The pharmacodynamic results weren't distinct different between the aqueous extract with alcohol precipitation and the aqueous extract:the effect on contraction of isolated rat uterine strips induced by oxytocin was increased significantly(P<0.01),as well as the case of vaginal opening of the premature female mice,the uterus weight percentages of the ovariectomized mice,and the number of ovulation of the mature mice after PMSG treatments.The optimized preparation techniques was adding 10 times water,extracting 1.0 h for twice,decompressing and concentrating the filtrate into relative density of 1.10(55℃),75% alcohol concentration was used for precipitation with in 24 h,concentrating and evaporating.Conclusion:The purposes of removing impurity,decreasing oral dosage and increasing therapeutic effects were achieved through water extraction and alcohol precipitation.The optimum preparation techniques was simple,reasonable,stable and useful for further development.
