Rho和ROCK蛋白在上皮性卵巢癌中的表达及临床意义

摘    要:目的探讨上皮性卵巢癌Rho和ROCK蛋白的表达及其与上皮性卵巢癌进展之间的关系。方法应用Western blot方法,观察了66例上皮性卵巢癌和22例正常卵巢组织Rho和ROCK蛋白的表达,并分析Rho/ROCK蛋白表达与生存期的关系。结果RhoA、C和ROCK蛋白在卵巢癌组织中表达增加,且在伴有肿瘤转移的淋巴组织中表达高于没有肿瘤转移的淋巴组织(P<0.05)。RhoA、C和ROCK间的表达呈正相关(P<0.05),RhoA、C和ROCK的表达与肿瘤分化、淋巴转移和短综合生存期相关(P<0.05)。多变量分析显示,只有RhoC是影响生存的独立因素(P<0.05)。RhoA和C是影响综合生存期的因素(P<0.05)。结论在上皮性卵巢癌中,RhoA、C和ROCK蛋白高表达,Rho/ROCK通路可能参与了上皮性卵巢癌的进展。

Abstract:Objective To investigate the expression of Rho and ROCK in epithelial ovarian carcinoma, and their relationship whih the progression of epithelial ovarian carcinoma. Methods Using Western blotting, we quantitated Rho and ROCK protein expression in 66 tumor samples from consecutive Chinese patients with epithelial ovarian carcinoma and 22 normal ovarian surgical samples without epithelial ovarian carcinoma. Using Immunohischemistry, we investigated the grades of tumors.Results Rho A, RhoC, and ROCK were more abundant in tumors and metastatic lymph nodes than in normal ovaries and unrelcted lymph nodes(P<0.05). The amounts of RhoA, RhoC, and ROCK protein expression correlated positively with one another(P<0.05). High RhoA, RhoC, and ROCK expression were related to poor tumor differentiation (P<0.05), and lymph node metastasis (P<0.05). Kaplan-Meier plots linked high RhoA, RhoC, and ROCK protein expression to shortened disease-free and overall survival(P<0.05). By multivariate analysis, only RhoC independently influenced disease-free survival(P<0.05),while RhoA and RhoC influenced overall survival(P<0.05). In contrast, RhoB expression was inversely related to the grade and stage(P<0.05), and its higher expression was associated with better overall survival(P<0.05). Conclusion The proteins of Rho A, RhoC, and ROCK were highly expressed in epithelial ovarian carcinoma. The Rho/ROCK pathway maybe involved in progression of epithelial ovarian carcinoma.
