坐骨神经损伤后脊髓前角运动神经元死亡性质的探讨

摘    要:目的:研究大鼠坐骨神经损伤后,脊髓前角运动神经元的死亡性质,以便加以保护,促进神经损伤后运动功能的恢复。方法:切断大鼠右侧坐骨神经,再原位吻合,于术后不同时间取L_4~L_6节段脊髓,作石蜡切片,通过H·E染色和Hoechst33258染色,光镜下分别观察前角运动神经元胞体的形态学变化特征;作超薄切片,电镜下观察前角运动神经元胞体超微结构的变化规律。结果:光镜下,右侧脊髓前角运动神经元胞体尼氏体和细胞核染色质浓缩;电镜下,细胞膜内陷,将细胞分割成凋亡小体,然后裂解,细胞体消失;而左侧前角运动神经元胞体均一、无变化。结论:坐骨神经损伤后,脊髓前角运动神经元有死亡,死亡性质是细胞凋亡。

Abstract:Functional restoration of a peripheral nerve following axotomy is unsatisfactoiy and axotomy-inducedneuronal cell death is believed to be one of the main reasons. Nerve growth factor(NGF) can protect spinal cordmotoncurons from axotomy-induced cell death. The character of the cell death is undefmite.The study demonstrates that the neuronal cell death is apoptosis after rat sciatic nerve injury.
