IL-1受体拮抗剂对培养人肾小管细胞的影响及机制研究

摘    要:目的:研究IL-1受体拮抗剂(IL-1ra)抗肾缺血再灌流性兔血清致培养人肾小管细胞损伤的作用及机制。方法:健康家兔16只,随机分为对照和缺血再灌流(IR)组,IR组作双肾动脉夹闭1小时再灌流2小时;对照组仅模拟实验过程,不夹闭肾动脉。分别提取其血清。肾小管细胞培养分对照组(DMEM+小牛血清);SC组(DMEM+对照兔血清);SIR组(DMEM+IR肾损伤性兔血清);IL-1ra+SIR组(DMEM+IR肾损伤性兔血清+IL-1ra)。培养96小时时,测定其增殖状况和细胞存活率,细胞或培养液中MDA、SOD、NO、LDH、Na~+-K~+-ATP酶和Ca~(2+)-ATP酶。结果:SIR组MDA、NO含量、LDH活力比对照组、SC组高(P<0.05),对照组、SC组和IL-1ra+SIR组间比较差异无显著性;肾小管细胞中的SOD活力,对照组最高,IL-1ra+SIR组次之,SC组又次之,SIR组最低,除IL-1ra+SIR组与SC组外,其余组间差异均有显著性(P<0.05);Na~+K~+-ATP酶和Ca~(2+)-ATP酶的活力在SIR组均较对照组和SC组低,差异有显著性。对照组、SC组和IL-1ra+SIR组间差异无显著性。结论:IL-1ra具有减轻IR性肾损伤兔血清致培养人肾小管细胞损伤的作用。

Abstract:Objective: To study the effect of IL- Ira on reducing the tubular cell injury caused by the serum from rabbit of renal ischemic repeifusion (1R) and its mechanism. Methods: (1)Serum preparation: 16 health rabbits were divided randomly into control and IR group. In IR group the bilateral renal arteries were occluded Ihour then reperfused 2hours, and the serum of control (SC) and of IR (SIR) were obtained. (2)Experiments on cultured human renal tubular cells: 4 groups were divided. Control group: The culture medium was DMEM plus calf serum; SC group: The culture medium was DMEM plus SC; SIR group: The culture medium was DMEM plus SIR; IL- Jra plus SIR group: The culture medium was DMEM plus IL - Ira (50ng. ml-1) and SIR. The content of MDA and NO, the activity of SOD and LDH and ATPase in the cell or culture medium were detected. The rate of living cells was assessed. Results: Compared with control and SC and IL- Ira + SIR groups, there was a remarkable increase in content of MDA in culture medium of SIR group. The content of NO and the activity of LDH in cell in SIR group were much higher than those in control group, SC group and IL- Ira + SIR group. But there was no significant difference in the content of MDA in culture medium and in the content of NO and in activity of LDH in cell among control and SC and IL - 1 ra + SIR group. The activity of SOD in cell was found highest in control group, next in IL - Ira + SIR group,and lowest in SIR group. There were significant differences between different group, except IL- Ira + SIR and SC group. The activity of Na+ - K4 - ATPase and Ca - ATPase in tubular cell in SIR group were markedly decreased compared with control group, SC group and IL - Ira + SIR group. But there were no remarkable differences in the activity of Na+ - K+ - ATPase and Ca2+ - ATPase in tubular cell among the latter three groups. Conclusion: IL - Ira could reduce renal tubular cell injury caused by the rabbit serum from acute renal ischemic reperfusion.
