摘    要:目的研究促性腺激素释放激素(GnRH)激动剂(GnRH agonist,GnRHa)对In-R1-G9细胞内胰升血糖素表达的调控作用。方法分别用终浓度为0.1nmol/L、10nmol/L、1000nmol/L的GnRHa处理培养的In-R1—09细胞2h(对照组用生理盐水处理),通过Western blot和荧光定量PCR技术观察胰升血糖素在蛋白水平及转录水平的变化。结果与对照组相比,低浓度的GnRHa(0.1nmol/L)使胰升血糖素在蛋白水平和转录水平的表达均有所升高,但差异无统计学意义(P〉0.05);而高浓度的GnRHa(10nmol/L、1000nmol/L)能明显增强胰升血糖素的表达(P〈0.05或P〈0.01)。结论一定浓度的GnRHa能增强In—R1-G9细胞内胰升血糖素的表达。

GnRH agonist may increase glucagon gene expression in In-R1-G9 cells

Abstract:Objective To study the effect of GnRH agonist (GnRHa) on the expression of glucagon in In-R1- G9 cells. Methods GnRHa was added to the media of the cells at final concemrations of 0. 1, 10, 1000 nmol/Lol/L, and normal saline added as control. At the end of the incubation, expressions of protein and mRNA of glucagon in In- RI-G9 cells were measured by Western blot and real time fluorescence/:~R respectively. Results /ks compared with control, high concentrations (10 nmol/L and 1000 nmol/L) of the GnRHa showed a significamly increased expressions of glucagon (all P〈0. 05) while low concentration (0. 1 nmol/L) did not show any significant charge (all P〈0. 05). Conclusion GnRHa at 10--1000 mnol/L may increase the expressions of glucagon in In-R1-G9 cells.
