磺脲类药物对胰岛β细胞凋亡的影响

摘    要:目的观察第二代和第三代磺脲类药物对胰岛β细胞INS-1凋亡的影响。方法采用大鼠胰岛素瘤细胞INS-1作为胰岛β细胞模型,药物干预后观察细胞形态学变化,用MTT和TUNEL法检测细胞增殖、凋亡情况,并测定基础和高糖刺激后胰岛素(Ins)分泌量。结果与阴性对照组相比,药物干预细胞4h,低浓度格列美脲对细胞凋亡无影响(P〉0.05),其他药物组细胞凋亡率明显增加(P〈0.05);药物干预1d及4d,各药物组细胞凋亡率明显增加(P〈0.05)。结论磺脲类药物可以促进胰岛β细胞的凋亡,且呈时间一剂量依赖性增加,提示对2型糖尿病(T2DM)患者临床治疗时要合理用药。

The effect of sulfonylureas on apoptosis of pancreatic beta cells of INS-1 strain

Abstract:Objective To study the effect of sulfonylureas on apoptosis of INS-1 cell. Methods INS-1 cells were routinely cultured and then the proliferation and apoptosis of the cells were evaluated by MTT and TUNEL, The insulin secretion was investigated by radioimmunoassay(RIA). Results As compared with negtive control group, after treatment for 4 hours, glimepiride didn't increase apoptotic cells (P〉0. 05); while apoptotic rate was increased in other drugs group (P〈0. 05). After treatment for 1 day, apoptotic cells increased obviously(P〈0. 05) ; after treatment for 4 days, apoptotic rate was much higher (P〈0. 05). Conclusion Sulfonytureas can increase apoptosis of INS-1 cells in a time-and dosedependent manner. This provides a new thought on studying pancreatic islet cell function and on reasonable use of sulfonylureas in patients with type 2 diabetes metlitus.
