体外循环对大鼠血脑屏障及紧密连接蛋白表达的影响

摘    要:目的探讨体外循环(CPB)对大鼠血脑屏障(BBB)功能及脑血管内皮紧密连接蛋白表达的影响。方法 24只成年雄性SD大鼠,随机分为假手术组(S组)和体外循环组(C组)。S组进行相应的麻醉和手术操作但不进行CPB,C组进行CPB 60 min并继续观察2 h。每组随机取6只测脑组织伊文思蓝(EB)含量,另6只做脑组织含水量测定、紧密连接蛋白Oc-cludin和ZO-1的表达以及大鼠BBB超微结构的观察。结果与S组比较,C组大鼠海马区脑组织EB含量和含水量升高,差异均有统计学意义(P〈0.05)。与S组比较,C组大鼠脑血管内皮紧密连接蛋白Occludin和ZO-1表达水平下降,差异均有统计学意义(P〈0.05)。S组BBB基膜均匀一致,完整性良好,内皮细胞间的紧密连接完整;C组基底膜变厚,微血管腔狭窄,内皮细胞间紧密连接不清楚。结论 CPB使大鼠BBB的结构发生变化、通透性增加,其机制可能与大鼠脑血管内皮紧密连接蛋白改变有关。
