| 摘 要： | 目的探讨miR-124对胃癌细胞MKN-45侵袭能力的影响。方法使用脂质体将miR-124mimics转染MKN-45细胞．以无关序列（scramblemimics）作为阴性对照。采用细胞划痕实验观察miR-124过表达对胃癌细胞MKN-45侵袭能力的影响。结果细胞划痕实验显示，转染miR-124mimics24h、48h后伤IZl愈合率分别为（0．313±0．005）、（0．369±0．031）．与各自对照组（0．413±0．035）、（0．852±0.024）比较能明显减缓MKN-45细胞划痕伤口的愈合，差异有统计学意义（P〈0．05）。结论miR-124能抑制胃癌细胞侵袭能力，它可能在胃癌的发生和进展中发挥重要作用。

| Abstract: | Objective To explicit whether miR-124 suppresses cell invasion and metastasis, thus to reveal molecular mech- anism that miR-124 functions as a tumorsuppressor in gastric carcinoma. Methods MKN-45 cells were transfected with miR-124 mimics, and next cell wound healing assay was performed to detect the invasion of miR-124. Results Transfection of miR-124 mimics for 24h, 48 h respectively, cell wound healing assay showed that the wound healing rate was （0.313±0.005） and （0.369±0.031）, compared with the control group （0.413±0.035） and （0.852±0.024）, it can＇ significantly slow down the scratch wound healing of the MKN-45 cell, the groups were significant different （P 〈 0.05）; Conclusion miR-124 can inhibit the invasion and metastasis in MKN-45 cells of gastric cacinoma. |
