帕金森病患者血淋巴细胞多巴胺转运蛋白功能与密度的研究

摘    要:目的:研究帕金森病(PD)患多巴胺转运蛋白(DAT)功能与密度的变化及其在:PD早期诊断中的意义。方法:实验对象分为PD组、正常老年对照组和脑血管病组。用放射性核素方法测定3组患的血淋巴细胞对^3H-多巴胺(DA)和^3H-WIN35428的放射性摄取率。结果:PD组的外周血淋巴细胞对^3H-DA和^3H-WIN35428的放射性摄取率低于脑血管病组和正常老年对照组;左旋多巴治疗的PD患外周血淋巴细胞对^3H-DA的放射性摄取率高于未治疗的PD患.对^3H-WIN35428的放射性摄取率两差异无显性。DA受体激动剂治疗的:PD患外周血淋巴细胞对^3H-DA和^3H-WIN35428的放射性摄取率均无影响。结论:PD患血淋巴细胞DAT的功能与密度明显降低,有可能作为疾病早期诊断的生物学指标。左旋多巴治疗对DAT的功能有促进作用但对其密度无影响;DA受体激动剂对DAT的功能和密度均无影响。

Abstract:Objective To investigate the function and density of dopamine transporter (DAT) in peripheral blood lymphocytes of patients with Parkinson's disease(PD) and to evaluate its role in early diagnosis of PD. Methods The uptake of 3H-dopamine and 3H-WIN35428 was measured by method of isotope in peripheral blood lymphocytes of patients with PD and stroke and healthy aged people. Results The uptake of 3H-dopamine and 3H-WIN35428 in blood lymphocytes of PD patients (n=42) was lower than that of healthy aged people (n=19) and stroke patients(n=26), regardless of the parkinsonian patients who received anti-Parkinson medication or not. Levodopa medication increased the uptake of 3H-dopamine in patients with PD, but had no influence on the uptake of 3H-WIN35428. No alternation of 3H-dopamine and 3H-WIN35428 uptake was found in patients with PD after administration of dopamine receptor agonist. Conclusion The function and density of DAT is significantly reduced in peripheral blood lymphocytes of parkinsonian patients, which might be a biological marker for the early diagnosis of Parkinson's disease. Levodopa medication improves the function of DAT in PD patients but not the density of DAT. Dopamine receptor agonist has no influence on the function and density of DAT in PD patients.
