| 摘 要： | 甲磺酸艾日布林(eribulin mesylate)是天然化合物软海绵素经结构优化后的大环酮类似物(代号E7389,ER-08652等),体内外试验结果显示其具有亚纳摩尔水平的抗肿瘤活性。2010年11月,甲磺酸艾日布林被美国FDA批准用于临床治疗至少接受过2种化疗方案(含蒽环类和紫杉类化疗药物)的转移性乳腺癌患者。本品商品名为HalavenTM,由Eisai公司开发上市。文中对其作用机制、药代动力学特性、I～III期临床研究结果以及安全性等进行综述。

| Abstract: | Eribulin mesylate(E7389,ER-08652) is a structurally optimized macrocyclic ketone analogue of halichondrin B,a natural compound,and has demonstrated promising anticancer activity in vitro and in vivo.US FDA has approved eribulin mesylate for patients with metastatic breast cancer who have failed at least two anthracycline-and taxane-based chemotherapy regimens.It was developed by Eisai Co.with the trade name of HalavenTM.This article summarizes the mechanism,pharmacology and the phase I,II,III clinical trials of eribulin mesylate. |
