中药"敛疡散"对慢性皮肤溃疡的作用机制研究

摘    要:目的:探讨敛疡散对慢性皮肤溃疡的作用机制。方法:采用敛疡散加入肉汤培养基中研究最低抑菌浓度;正常豚鼠金黄色葡萄球菌感染引起的皮肤疮疡及环磷酰胺处理的豚鼠疮疡模型,比较敛疡散和黄连膏治疗后疮疡的红肿、分泌物、疮口的改善情况以及脓液中溶菌酶的水平;利用热板法和化学法观察敛疡散对小鼠痛阈改变;观察敛疡散对小鼠耳肿胀、大鼠毛细血管通透性及大鼠琼脂肉芽肿作用;观察敛疡散对巨噬细胞百分率和吞噬指数改变以及对小鼠鸡红细胞攻击的小鼠的溶血素及环磷酰胺处理的小鼠溶血素作用。结果:敛疡散对金黄色葡萄球菌、绿脓杆菌、大肠杆菌的最低抑菌浓度分别为0.00547g/ml,0.02188g/ml,0.01094g/ml;敛疡散对正常豚鼠金黄色葡萄球菌感染引起的皮肤疮疡及环磷酰胺处理的豚鼠疮疡模型中疮疡的红肿、分泌物、疮口得到良好的改善,脓液中溶菌酶的水平治疗后也的明显的提高,而且敛疡散的作用比黄连膏对照有显著性差异(P<0.01);物理法和化学法镇痛实验均证明敛疡散对小鼠的痛阈具有明显的提高作用,特别是大剂量组的作用更为明显;敛疡散对小鼠耳肿胀、大鼠毛细血管通透性及大鼠琼脂肉芽肿均表现出非常明显的抑制作用。敛疡散对实验动物的特异性免疫和非特异性免疫功能均具有增强作用,表现为巨噬细胞百分率和吞噬指数均明显地提高,作用强度呈剂量依赖性,且作用强度明显优于黄连膏对照组(P<0.01);对小鼠鸡红细胞攻击的小鼠的溶血素及环磷酰胺处理的小鼠溶血素均具有一定程度的提高作用,且作用优于黄连膏对照组。结论:敛疡散具有抗菌抑菌作用,抗实验性疮疡作用,明显的抗炎、抗渗出、抗增生、镇痛作用,以及对机体免疫功能具有调节作用,因此对慢性皮肤溃疡有良好的疗效。

Abstract:Objective: To study skin ulcer pharmacodynamics of Lianyangsan(LYS). Methods: To observe the minimum inhibitory concentration(MIC); to construct guinea pigs models of skin ulcer were established by inducing infection and cyclophosphamide, comparing with Huangliangao(HLG), the condition of uncle was in investigated; variation of pain threshold was observed for two kind of physical and chemical method; change of mice ear oedama and permeability of capillary on rat skin function was picked up; then the effect of LYS on phagocytosis of mouse enterocoelia macrophage was evaluated by the chicken-red cells phagocytosis index and phagocytosis rate. Results:They were found to be active against the four test bacterial organisms; dimelioration of skin ulcer was found including colour,swelling and secretion; abirritation on small dose group and large dose group are evidently greater than that of the group applying HLG; to impact on permeability of capillary on rat skin function of LYS is significantly greater than that of HLG; tumefaction control rate of the group applying LYS is higher than that of positive control group; the phagocytosis index and phagocytosis rate of experimental groups both enhance. Conclusions: LYS has better functions of antiinflammation, abirritation ,detumescence as well as improvement of skin ulcer and immunomodulation. antimicrobial activity
