摘    要:目的:采用RP-HPLC对蒲公英叶及根中咖啡酸的含量进行测定。方法:选用Hyper ODS C18色谱柱(250mm×4.6 mm),甲醇-磷酸盐缓冲液(23∶77)为流动相,紫外检测波长为323 nm,柱温:40℃条件下对咖啡酸进行含量测定。结果:咖啡酸的含量在0.145μg~0.726μg范围内,进样量与峰面积积分值呈良好的线性关系,r=0.9996,平均回收率98.3%,RSD=1.80%。结论:此方法能准确、快速地进行定量检测,可作为蒲公英叶及根中咖啡酸的质量控制方法。

Abstract:Objective: To develop a RP-HPLC method for the caffeic acid in leaves and roots of Herba taraxacum mongolicum,which determined the quantity of caffeic acid.Methods: The chromatographic conditions included Hyper ODS C18 column(250 mm×4.6 mm) and the mobile phase consisted of methanol-phosphate buffer solution(23∶77).The detection wavelength was at 323 nm.Results: Caffeic acid showed a good linear relation ship at the rang of 0.145 μg~0.726 μg,r = 0.9996.The average recovery rate was 98.3% and RSD was 1.80%.Conclusion: The methods are accurate and quick,and can be used for the quality control of caffeic acid in leaves and roots of Herba taraxacum mongolicum.
