人端粒保护蛋白的研究进展

摘    要:人端粒保护蛋白(human protection of telomeres,hPOT1)是一种端粒单链DNA结合蛋白,它结合于端粒末端的单链重复序列,在端粒长度的调控以及染色体的稳定方面起重要的作用。其表达水平下降所引起的端粒功能异常以及染色体畸变,与人类细胞的老化和恶性肿瘤的发生发展有密切的关系。对hPOT1结构、功能及作用机制的研究,不但可以进一步阐明细胞衰老和恶性肿瘤发生的分子机制,而且也将为肿瘤的诊断、治疗开辟新的思路和方法。

Abstract:hPOT1 (human protection of telomeres, hPOT1) is a kind of telomeric single-strand DNA binding protein, which binds to the most distal single-stranded extension of telomeric DNA in vivo and is proposed to play an important role in the regulation of telomere length and stability of chromosome.Decreased hPOT1 expression may cause telomere dysfunction and chromosomal aberration, which have been closely implicated in human aging and cancer.Therefore, study of the structure, functions and action mechanisms of hPOT1 not only contributes to the understanding of the molecular mechanisms of cell aging and tumorogenesis, but also helps to develop new techniques of tumor diagnosis and treatment.This article reviews the advances in the research on hPOT1.
