fGenistein 后处理对缺血再灌注大鼠SP-A的影响

摘    要:摘要:目的 观察 Genistein 药物后处理对缺血再灌注损伤(IRI)大鼠肺表面活性物质相关蛋白A的影响,并探讨该方法对肺缺血再灌注损伤的作用机制。方法 32只雄性SD大鼠随机分为空白对照组(Sham)、缺血再灌注组(IR)、缺血后处理组(IPO)、药物后处理组(Genistein),每组8只。测定肺组织湿/干重比(W/D)、超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮合酶(NOS)、肺表面活性物质相关蛋白A(SP-A)含量,并在光镜下观察各实验组肺组织结构变化,结果 与IR组相比较,药物后处理组W/D、MDA、iNOS/T-NOS比值显著降低(P〈0.05),SOD、SP-A显著升高(P〈0.05)。而药物后处理组和缺血后处理组的各项指标均无明显差异(P〉0.05)。光镜下观察可见缺血后处理组和药物后处理组肺泡和支气管壁充血水肿较缺血再灌注组明显减轻,与空白对照组较为接近。结论 Genistein药物后处理对肺缺血再灌注损伤有保护作用,效果与缺血后处理相似,可能与灭活氧自由基,减少sP-A的损失有关。

Abstract:Objective To study the effective of C, enistein Postconditioning on lung ischemia-reperfusion injury and the Influence on SP-A. Methods A total of 32 male SD rats were randomly divided into 4 groups,8 rats in each group. The W/D, SOD, MDA, NOS, SP-A in lung tissue were detected, and the histological changes of lung was observed under light microscope. Results Compared with IR group, the W/D, MDA and iNOS/TNOS were significantly reduced in genistein group, and SOD, SP-A were significantly increased. There was no significant difference between IPO group and genistein group. Under light microscope, the hyperemia and edma of alveolar and bronchus epithelial in IPO and genistein groups were obviously alleviated as compared to IR group. Conclusion C, enistein postconditioning has important protective effect on lung ischemia-reperfusion injury, which is similar with IPO, and the mechanism may be involve in the scavenging of oxygen species and reduction of SP-A loss.
