摘    要:目的:克隆人类抗凋亡基因survivin(SVV)。方法:提取胃癌细胞系SGC-7901总RNA,RT-PCR扩增人SVV全长cDNA,测序鉴定、原核表达。结果:得到两个SVV基因cDNA克隆,即SVV-S4A与SVV-S1B,前者与已知SVV基因cDNA序列相同,后者第3个外显子丢失。原核表达所获融合蛋白经SDS-PAGE电泳与已知SVV基因的蛋白质表达产物大小相近。结论:自人胃癌细胞中克隆出SVV基因的全长cDNA,并获得一个新的剪切体,实现了SVV基因在大肠杆菌BL21(DE3)中的高效表达。

Abstract:Objective:The purpose of this paper is to clone survivin(SVV) gene from human gastric cancer. Methods: The SVV cDNA was obtained by using RT PCR method with total RNA extracted from the human gastric cancer cell line, SGC7901. After proved to be correct by sequencing , recombinant expression plasmid p RSET SVV was transformed into E.coli BL21(DE3). The fusion protein was produced by IPTG induction. Results : Obtained two SVV cDNA clones , SVV S 4 A and SVV S 1 B. The sequence of the previous is identical to that of the well known SVV, but , the third exon was missed in the latter.A new anticipated M r 19 500 protein band appeared on SDS PAGE gel induced by IPTG. Conclusion: human SVV cDNA was cloned and a new splicer is found from human gastric cancer. SVV is highly expressedsioned in E. coli.
