小鼠毛囊来源间充质干细胞体外诱导成骨的实验研究

摘    要:目的:探索便捷而有效的小鼠毛囊来源间充质干细胞的体外培养方法,并研究该群细胞的成骨能力,以探寻新的骨组织工程种子细胞。方法:采用酶消化法分离获得小鼠毛囊来源的干细胞,用不同培养体系进行传代培养,观察各细胞的增殖能力,并对所获得的毛囊来源的干细胞向成骨细胞诱导,行碱性磷酸酶染色、茜素红染色,对该细胞的成骨能力进行鉴定。结果:应用MEF条件培养液培养的小鼠毛囊来源的间充质干细胞具有较强的增殖能力,且诱导组和对照组能在体外被诱导成骨。结论:小鼠毛囊中存在具有较强增殖能力及成骨能力的间充质干细胞,可作为骨组织工程的种子细胞。

Abstract:PURPOSE: To find an easy and efficient method of isolating and culturing mesenchymal stem cells derived from murine hair follicle in vitro,and evaluate the osteogenic differentiation potential and the possibility of being a new seed cell of bone tissue engineering of these cells.METHODS: Hair follicle stem cells were seperated from murine dorsal skin with enzyme digestion and cultured with 4 different mediums.The cell morphology and growth were observed by phase contrast microscopy.Then the selected cells were induced to osteoblasts.Production of calcified deposits were detected by alkaline phosphatase staining and Alizarin red staining.RESULTS: The primary cell line cultured with MEF conditioned medium had extended proliferative capabilities.Both the inductive group and control group was capable of being directed to bone differentiation.CONCLUSION: Our findings showed that within the hair follicles there really exist the mesenchymal stem cells which have considerable proliferative capabilities and could be used for seed stem cell in bone tissue engineering.
