摘    要:目的:采用高效液相色谱法(HPLC)测定辣椒粉中的碱性橙。方法:称取样品于50 ml塑料离心管中,加入10 ml提取液,超声、离心后,取上清液过膜进样。采用XDB-C18色谱柱(5μm,4.6×250 mm),流动相为乙腈+乙酸铵(75+25),流速1.0 ml/min,二极管阵列检测器检测。结果:碱性橙在1.0μg/ml~50.0μg/ml范围内线性良好(r=0.9992),且峰形较好,回收率为93.4%,精密度RSD=0.61%,最低检出限为0.3μg/ml。结论:高效液相色谱法测定辣椒粉中的碱性橙,方法定性准确、灵敏度高、回收率和重现性好,操作快速,结果准确可靠。

Abstract:Objective:To use high performance liquid chromatography(HPLC) method for determination of basic orange in paprika.Methods: Samples were weighed in 50 ml plastic centrifuge tube with 10 ml extraction.After ultrasonication and centrifugation,the supernatant was taken for filtration through the membrane and sample injection.The separations were performed with an XDB-C18 column(5 μm,4.6×250 mm) at flow rate of 1.0 ml/min.The mobile phase was acetonitrile-ammonium acetate(75+25).The diode array detector was applied.Results: The basic orange showed good linearity in the range of 1.0 μg/ml~50.0 μg/ml(r=0.9992) and better peak shape.The recoveries were 93.4% and the RSD was 0.61%.The detection limit was 0.3 μg/ml.Conclusion: The method for determination of basic orange by HPLC in paprika have the advantages of qualitative accuracy,high sensitivity,good recovery and reproducibility,fast operation,accurate and reliable results.
