青蒿素对糖尿病心肌病大鼠心功能及纤维化的影响

摘    要:目的观察青蒿素对糖尿病心肌病大鼠心功能及纤维化的影响。方法 Wistar大鼠随机取正常组8只,其余大鼠以一次性腹腔注射链脲佐菌素造糖尿病模型。16周后,经超声检测糖尿病心肌病造模成功大鼠,再随机分为糖尿病心肌病组、青蒿素治疗组,每组8只。各组分别采取相应干预措施。采用HE染色观察心脏的病理改变,用Masson三色染色法观察胶原纤维表达并测定其表达量,应用超声心动图评测Wistar大鼠的心脏功能。结果与正常组比较,糖尿病心肌病组胶原纤维的表达明显升高(P0.01),E/A、LVEF、FS明显下降(P0.01),心脏收缩舒张功能明显降低。与糖尿病心肌病组比较,青蒿素治疗组大鼠胶原纤维的表达明显降低(P0.01),E/A、LVEF、FS明显上升(P0.01),心脏收缩舒张功能明显改善。结论青蒿素可通过减轻心脏的纤维化来提高大鼠的心功能。

Abstract:  Objective:To observe the effects of artemisinin on cardiac function and fibrosis in diabetic cardiomyopathy rats. MethodsEight Wistar rats were randomly selected as normal group, and the other rats were created as diabetic models by intraperitoneal injection of streptozotocin. After 16 weeks, the diabetic models were set up successfully after the test, and were randomly divided into diabetic cardiomyopathy group and artemisinin group, 8 in each group. Each group was intervened respectively. The myocardial pathology structure was observed by HE staining. The content of collagen was detected by Masson staining, and cardiac function was measured by Echocardiography.  Results:Compared with normal group, the expression of fibrosis increased obviously in diabetic cardiomyopathy group(P<0.01), and E/A, LVEF and FS decreased obviously, and cardiac systolic and diastolic function decreased obviously. Compared with diabetic cardiomyopathy group, the expression of fibrosis decreased obviously(P<0.01), E/A, LVEF and FS increased(P<0.01), and cardiac systolic and diastolic function improved obviously.  Conclusion:Artemisinin may improve cardiac function of rats by attenuate myocardial fibrosis.
