体外培养人肝癌细胞系7721中HCV复制和表达特性

建立支持ＨＣＶ体外长期复制的细胞培养体系，研究其体外复制和表达特性。方法：将ＨＣＶ感染血清接种人肝癌细胞系７７２１后，以逆转录－聚合酶链反应、免疫组化、原位杂交检测细胞内ＨＣＶ的感染和复制情况。结果细胞内和培养上清中在孵育后的３月余均可间断地检测出ＨＣＶ正链和／或负链ＲＮＡ，多种ＨＣＶ抗原在细胞内能稳定有达；ＨＣＶ ＲＮＡ和抗原多位于胞浆中，结论ＨＣＶ在７７２１细胞系中的复制和表达与体内感染状态接

Characteristics of replication and expression of HCV cultured in a human liver cancer cell line in vitro

Objective To establish a long term cell culture system in order to study the characteristics of HCV replication and expression in vitro. Methods Human liver cancer cell line 7721 was incubated in HCV positive serum for 8 hours and then the infection and replication of HCV in 7721 cells were observed with RT PCR, in situ hybridization and immunohistochemistry. Results The plus and minus-strand of HCV RNA was intermittently detected in the cells and the supernatant of the culture medium over a period of more than three months after incubation. Various antigen against HCV were expressed in cells. HCV RNA and the antigens were mainly located in the cytoplasm. Conclusion Our findings suggest that the replication and expression of HCV in the liver cancer cell line are similar to HCV infection of human body and this long term cell culture system can serve as a valuable tool for the further study of HCV infection.
