摘    要:目的研究在促红细胞生成素(erythropoietin,EPO)和甲泼尼龙(Methylprednisolone,MPSS)干预下对损伤星形胶质细胞增殖的影响,探讨EPO和MPSS联合应用在脊髓损伤中的作用。方法取自SD大鼠大脑原代培养星形胶质细胞,缺营养培养3h后分别在促红细胞生成素(erythropoietin,EPO)和甲泼尼龙(erythropoietin,EPO)的培养基中培养24、48小时后,MTT法检测星形胶质细胞增殖活性的变化。结果缺营养可抑制星形胶质细胞增殖,促红细胞生成素和甲泼尼龙对正常细胞增殖无明显影响作用,EPO和MPSS均促进损伤细胞增殖,两者联合减量应用对细胞增殖有明显影响作用。结论 EPO+MPSS(半量组)联合减量应用对星形胶质细胞增殖作用表达的升高作用优于单独应用甲泼尼龙。

The effect of erythropoietin and methylprednisolone in combination on astrocytes

Abstract:Objective To study the effect of erythropoietin(rhEPO) and methylprednisolone(MPSS) in combination on ischemic injuried astrocytes(AST) in vitro culture.Methods The astrocytes were isolated from cerebral cortex of neonatal rats born within three days,cultivating by DMEM media added with 15% fetal calf serum and depurating by swinging and passaging,verificating astrocytes by immunofluorescent technique.To simulate ischemic model by substituting medium for PBS buffer solution 3h then applying erythropoietin(10U/ml),methylprednisolone(10μg/ml),erythropoietin(10U/ml)+methylprednisolone(10μg/ml),erythropoietin(5U/ml) +methylprednisolone(5μg/ml)and DMEM media added with 15% fetal calf serum immediately,then went on culturing,detecting activity of Ast by MTT.Results Hypso-purity(95%)astrocytes could be acquired and more depurative by passaging,part of astrocytes will be dying after ischemic;EPO and methylprednisolone had obviously protection effect for damaged astrocytes.Conclusion The applying of half dose of EPO and methylprednisolone have obviously protection effect for damaged astrocytes.
