氯丙烯对鸡胚脑神经细胞钙稳态与能量合成影响的研究

摘    要:进一步研究氯丙烯的中毒机理,探讨氯丙烯对神经细胞内钙稳态与能量合成的影响。方法用Fura-2/AM、电子探针X线微区分析、ATP生物发光分析等方法测定氯丙烯染毒后鸡胚脑神经细胞内游离Ca2+、钙百分含量及ATP、ADP、AMP含量的改变。 结果随着氯丙烯浓度的增加,细胞内游离Ca2+浓度、钙百分含量明显增加(P

Abstract:Objective: To research the poisoning mechanisms of allyl chloride and probe into the action of allyl chloride on calcium homeostasis and energy synthesis in brain cells.Methods: Contents of Ca, ATP, ADP, AMP were studied in allyl chloride induced chicken embryo brain cells with the methods of fluorescence molecular probe Fura 2/AM, electron probe X ray microanalysis(EXMA)and ATP bioluminescence analysis. Results: With the increase of allyl chloride, the contents of Ca 2 increased significantly (P<0.01), while the contents of ATP, ADP, AMP markedly decreased(P<0.01).Conclusion: The results suggested that allyl chloride can increase the concents of calicium and decrease energy synthese in nerve cells. Inhibition of energy synthesis in duced by allyl chloride in nerve cells might be one of the mechanisms of calcium unhomeostasis.
