摘    要:目的:研究二甲亚砜诱导人胃腺癌MGC803细胞分化及其AKPase表达。方法:分别提取诱导前后胃腺癌MGC803细胞、胎盘、H4移植瘤碱性磷酸酶,并通过神经氨酸酶处理后聚丙烯酰胺凝胶电泳分析及敏感实验(苯丙氨酸、温度、咪唑对AKPase活性影响)作AKPase特性检测。MTT法,酶荧光染色法研究胃腺癌MGC803细胞经诱导后细胞生长及AKPase表达,结果:诱导前后胃腺癌MGC803细胞均表达两型

Abstract:Objective:To search the differential effects of DMSO on human gastric adenocarcinoma cell line MGC803 and changes of its alkaline phosphatase expression. Methods: Alkaline phosphatase was extracted from human gastric adenocarcinoma cell line MGC803 induced 1.5% DMSO 7days or not,placenta,liver,H4 hepatoma tissue, neuraminialase treatment and then subjected to polyacrylamide gel disc electrophoresis.Some of the extract was used for analying the extent of inhibition by amino acid,heat stability,imidezole.Cultured MGC803 cell multiplication and AKPase quantity were analysed,contrasted with induced cell. Results: there were two types of AKPase inducing or not expressing in MGC803.The inhibition analysis showed AKPase in MGC803 was similar to placental-type while treated cell was similar to live-type. Induced MGC803 cell multiplication decreased to 66% and AKPase quantity/cell decreased to 68%. Conclusion: DMSO had differential effect on human gastric adenocarcinoma cell line MGC803 and changes the alkaline phosphatase expression.
